It's a simple,You just have to blog the object which one you want to fill with
color or if it all of image you can push CTRL+A then push CTRL+ALT+X
and now you can use paint bucklet or just push 'K'
that is for Basic color
If you want create a shading you just have to create more vector to that
shadow
if that it finish you can click in the Swatch color what you want

but I recommend to use Illustrator
because if you use photoshop that is not an vector but BMP
if you want to draw you can use pen tool
or P in shortcut
just try to make one
you can follow the line with click in the anchor point and I think if you try
you will understand by yourself

I capture my work in Illustrator like your request,I think it will be many cap.

actually I want to create something like video tutorial with adobe capative
but the result is not good
because the picture is very small and I think you can't watch something like that.hehehehe

BTW I not use a pen mouse just a normal mouse
because to create vector you don't need to have a pen mouse
except you want to use Photoshop for brush or manual draw in photoshop
mmmm,now my class have a plan to watch 'Hanami' together
